AKTIVITAS ENZIM PROTEASE DAN KONDISI PENCERNAAN DI USUS IKAN KERAPU MACAM (Epinephelus fuscoguttatus) SETELAH PEMBERIAN PAKAN

Abstract


Penelitian ini bertujuan untuk melihat aktivitas enzim protease dan kondisi makanan di usus ikan kerapu macan (Epinephelus fuscoguttatus) setelah pemberian pakan. Penelitian dilakukan di keramba jaring apung (KJA) Balai Riset Perikanan Budidaya Air Payau di Teluk Awarange Kabupaten Barru, Sulawesi Selatan. Ikan yang digunakan berukuran 200 ± 10 g berumur 6 bulan. Tujuh buah keramba berukuran 1 m x 1 m x1,2 m digunakan untuk memelihara ikan yang diletakkan secara acak untuk setiap pengambilan. Semua ikan diberi pakan moist pellet dengan kadar protein 45% sampai ikan kenyang (ad satiation). Sampling dilakukan pada 6, 9, 12, 15, 18, 21, dan 24 jam setelah pemberian pakan. Preparasi dan analisis aktivitas enzim protease dilakukan dengan pengambilan cairan usus sesaat setelah ikan dimatikan. Aktivitas protease diukur secara kuantitatif dengan modifikasi metode Bergmeyer. Hasil analisis enzim protease menunjukkan bahwa aktivitas tertinggi diperoleh pada 18 dan 21 jam setelah makan (JSM) yaitu 13,04 μ /mL dan 13,3 μ/mL dan terendah pada 9 JSM yaitu 5,83 μ/ mL. Hasil analisis persentase cairan usus menunjukkan hasil yang fluktuatif antara tiap jam pengamatan. Persentase cairan usus tertinggi diperoleh pada 9 JSM yaitu 76% dan terendah pada 6 JSM yaitu 53%.
Aims of the research are to figure out proteolitic activities and intestinal digestibility on tiger grouper (Epinephelus fuscoguttatus) after feeding. The research was carried out on Research Institute for Coastal Aquaculture (RICA) floating net cages at Awarange Bay, Barru Regency, South Sulawesi.The fish has 200 ± 10 g on size and relatively same on age of 6 months. Seven floating net cages of 1x1x1.2 m3 were used to rearing fish with random position for each sampling. The fish fed with 45% protein of moist pellet ad satiation. Sampling was carried out at 6, 9, 12, 15, 18, 21, and 24 after fed. Preparation and analysis of protease enzyme activity collected after grouper was killed. Protease activity was analyzed by modification of Bergmeyer methods. Result of enzyme analysis show that 18 and 21 hours after fed has the highest activities i.e. 13.04 μ/mL and 13.3 μ/mL respectively, and 9 hours after fed has the lowest activities i.e. 5.83 μ/mL. Result of liquid presentation of intestinal shows fluctuation in every time of sampling. The highest presentation of liquid was at 9 hours after fed of 76% and the lowest was 6 hours after fed of 53%.
